- LiP (lignin peroxidase) from Trametopsis cervina has an exposed catalytic tyrosine residue (Tyr181) instead of the tryptophan conserved in other lignin-degrading peroxidases. Pristine LiP showed a lag period in VA (veratryl alcohol) oxidation. However, VA-LiP (LiP after treatment with H2O2 and VA) lacked this lag, and H2O2-LiP (H2O2-treated LiP) was inactive. MS analyses revealed that VA-LiP includes one VA molecule covalently bound to the side chain of Tyr181, whereas H2O2-LiP contains a hydroxylated Tyr181. No adduct is formed in the Y171N variant. Molecular docking showed that VA...AuthorsYuta Miki, Rebecca Pogni, Sandra Acebes, Fatima Lucas, Elena Fernandez-Fueyo, Maria Camilla Baratto, Maria I. Fernandez, Vivian De Los Rios, Francisco J. - This study evaluated the potential effectiveness of future carbon reserve scenarios, where U.S. forest landowners would hypothetically be paid to sequester carbon on their timberland and forego timber harvests for 100 years. Scenarios featured direct payments to landowners of $0 (baseline), $5, $10, or $15 per metric ton of additional forest carbon sequestered on the set aside lands, with maximum annual expenditures of $3 billion. Results indicated that from 1513 to 6837 Tg (Teragrams) of additional carbon (as carbondioxide equivalent, CO2e) would be sequestered on U.S. timber-lands...AuthorsPrakash Nepal, Peter J. - Comparisons of properties are made for pressure-sensitive adhesives (PSAs) generated via emulsion polymerization using both conventional and reactive emulsifiers. The emulsifiers are ammonium salts of sulfated nonylphenol ethoxylates with similar chemical structures and hydrophilic−lipophilic balances. The polymerizable surfactant possesses a reactive double bond at its phenyl ring allowing for participation in free-radical polymerization reactions. Incorporation of the polymerizable surfactant into the adhesive polymer was confirmed via reverse-phase liquid chromatography. Surfactant...AuthorsJilin Zhang, Yuxi Zhao, Matthew R. Dubay, Steven J. - We used a new cellulosic material, cellulosic solid residue (CSR), to produce cellulose nanofibrils (CNF) for potential high value applications. Cellulose nanofibrils (CNF) were produced from CSR recovered from the hydrolysates (waste stream) of acid hydrolysis of a bleached Eucalyptus kraft pulp (BEP) to produce nanocrystals (CNC). Acid hydrolysis greatly facilitated homogenization to fibrillate CSR to CNF with only 15 passes in a microfluidizer compared with at least 47 passes to fibrillate BEP to nanofibrils. CNF from CSR were nanowhiskers with a length between 50 and 400 nm and a...AuthorsQianqian Wang, Junyong Zhu, John M. ConsidineKeywordsSourceACS Appl. Mater. - Fast-growing biomass, such as bamboo, has the potential to serve an important future role in the pulp and paper industry with potential to both lower resource costs and improve a product’s sustainability. Moso bamboo is particularly interesting due to its fast growth and size, which allows it to be handled and chipped similarly to wood resources. In this study, we will share results of the chip preparation, kraft cooking, and ECF bleaching of this bamboo species and compare its pulpability, bleachability, and physical properties to a fast growing hybrid poplar tree. Results indicate that...AuthorsTroy Runge, Carl J. Houtman, Alberto Negri, Jackie HeinricherKeywordsSourceTAPPl JOURNAL, Volume 12, Number 2 , FEBRUARY 2013; pp. 9-15.Year2013
- The oxidative enzymatic machinery for degradation of organic substrates in Agaricus bisporus (Ab) is at the core of the carbon recycling mechanisms in this fungus. To date, 156 genes have been tentatively identified as part of this oxidative enzymatic machinery, which includes 26 peroxidase encoding genes, nine copper radical oxidase [including three putative glyoxal oxidase-encoding genes (GLXs)], 12 laccases sensu stricto and 109 cytochrome P450 monooxygenases. Comparative analyses of these enzymes in Ab with those of the white-rot fungus, Phanerochaete chrysosporium, the brown-rot...AuthorsHarshavardhan Doddapaneni, Venkataramanan Subramanian, Bolei Fu, Daniel CullenKeywordsSourceFungal Genetics and Biology Volume 55, 2013; pp. 22-31.Year2013
- Basidiomycetes that cause brown rot of wood are essential biomass recyclers in coniferous forest ecosystems and a major cause of failure in wooden structures. Recent work indicates that distinct lineages of brown rot fungi have arisen independently from ligninolytic white rot ancestors via loss of lignocellulolytic enzymes. Brown rot thus proceeds without significant lignin removal, apparently beginning instead with oxidative attack on wood polymers by Fenton reagent produced when fungal hydroquinones or catechols reduce Fe3+ in colonized wood. Since there is little evidence that white...AuthorsPremsagar Korripally, Vitaliy I. Timokhin, Carl J. Houtman, Michael Mozuch, Kenneth E. HammelKeywordsSourceAppl. Environ. Microbiol. - When hydrogen peroxide is mixed with caustic soda, an auto-accelerating reaction can lead to generation of significant amounts of heat and oxygen. On the basis of experiments using typical pulp mill process concentration and temperatures, a relatively simple kinetic model has been developed. Evaluation of these model results reveals that hydrogen peroxide-caustic soda systems are extremely sensitive to hydrogen peroxide:caustic soda ratio, transition metal contamination, and temperature. Small changes in initial conditions can result in a close system becoming explosive. Analysis of model...AuthorsPeter W. Hart, Carl J. - The effectiveness of treatments for the surface layer of novel foam core particleboards was evaluated by means of Cone calorimeter tests, Foam cote particleboards with variations of surface layer treatment, adhesives, and surface layer thicknesses under similar processing conditions were used to produce the test specimen for the Cone calorimeter tests. Ignitability, heat release rate profile, peak of heat release rate, total heat released, effective heat of combustion, mass loss rate, gaseous emissions, and specific extinction area were measured using the cone irradiance of 50 kW m-2....AuthorsMark A. - Both cellulose nanocrystals (CNCs) and cellulose nanofibrils (CNFs) are nanoscale cellulose fibers that have shown reinforcing effects in polymer nanocomposites. CNCs and CNFs are different in shape, size and composition. This study systematically compared their morphologies, crystalline structure, dispersion properties in polyethylene oxide (PEO) matrix, interactions with matrix, and the resulting reinforcing effects on the matrix polymer. Transparent PEO/CNC and PEO/CNF nanocomposites comprising up to 10 wt % nanofibers were obtained via solution casting.
